3D Cryo-FIB/SEM image processing tool available

Our custom-made image processing tool for Cryogenic Focused Ion Beam / Scanning Electron Microscopy (Cryo-FIB/SEM) image stacks is now publicly available!3D Cryo-FIB/SEM generates 3D reconstructions of samples by cyclically milling a layer off the vitrified sample and then imaging the newly exposed surface. It combines the advantages of sample preservation at near-native fully-hydrated states and serial acquisition of nanometer-thin sections, enabling visualization of subcellular structures in large volumes with high resolution. However, resulting images can be riddled with artifacts. 

To address these issues, our Cryo-FIB/SEM Image Processing tool offers several functions for correcting (1) curtaining artifacts, (2) charging artifacts, (3) noise, (4) low contrast, (5) uneven brightness across slices of the stacks. Built within the napari framework, which offers a graphical user interface, the processing workflow is convenient and user-friendly from start to finish.
